Summary: I am mainly interested in the understanding of the signal transduction events that drive oncogenic cellular transformation. In particular my research projects are focused on the understanding of the causes of apoptotic resistance in the process of cellular transformation. It is well established that the acquired capability to escape apoptosis is required at several steps during cancer development. The possibility to re-establish normal sensitivity to apoptotic cell death is therefore a major task for effective cancer treatment. In this context, over-expression of members of the Bcl-2 family is known to play a croucial role. I'm therefore investigating both biochemical consequences of Bcl-xL increased expression, and the signal transduction pathways responsible for its expression. In this context I found a number of novel Bcl-xL interacting proteins that might shed light on the full phisyological significance of Bcl-xL overexpression in cancer progression. Another oncogene I'm interested in is c-Myc. Activation of c-Myc has been shown to be necessary but not sufficient for full transformation because of its inherent propensitiy to induce apoptosis. By establishing a non-transformed, telomerase-immortalised human epithelial cell line expressing an activatable version of the c-Myc protein, I have found that c-Myc activation, in addition to sensitisation to apoptosis, induces a marked reduction in cell motility and invasiveness. Interestingly, through the analysis of the genes regulated by c-Myc, I found that c-Myc caused a strong downregulation of uPA and its receptor uPAR. Interestingly, downregulation of the uPA/uPAR system seems to account for most of the proapoptotic and antimigratory effect exerted by c-Myc activation. The subsequent introduction of a constitutively active form of Ras can override both these tumour suppressor-like activities.
